En su ambición por dominar Japón, el cruel señor Daigo Kagemitsu hace un pacto terrible: sacrifica a su futuro hijo a 48 demonios. Aceptan, y el niño nace desmembrado, arrojado a un río para morir.
Quince años después, un joven huérfano llamado Dororo lucha por sobrevivir en un país devastado por la guerra. Tras un altercado, es salvado por un misterioso joven que empuña espadas ocultas en sus brazos protésicos. Este joven, Hyakkimaru, revela que su cuerpo fue despojado por demonios y que, aunque anhela la paz, no puede escapar de su destino.
A pesar de la dura verdad, Dororo decide unirse a Hyakkimaru en su viaje. Juntos, acompañados por un cachorro llamado Nota, recorren Japón enfrentando la crueldad de los demonios y la brutalidad de los hombres, en una búsqueda por recuperar lo que fue robado y encontrar un lugar en el mundo.
