En Magano, un mundo paralelo plagado de monstruos conocidos como "Kegare", los exorcistas se encargan de purificar toda impureza. Benio Adashino, una prodigio reconocida por su fuerza, es convocada a Tokio por la Unión de Exorcistas. En su camino, cae en brazos de Rokuro Enmadou, un joven exorcista atormentado por su pasado.
Pero las impurezas de Magano no dan tregua. Cuando ambos exorcistas presencian cómo un Kegare se lleva a unos niños, Benio se lanza a su rescate, arrastrando a Rokuro con ella a ese sombrío reino. En medio de una batalla donde Benio está a punto de ser derrotada, Rokuro interviene, demostrando ser un rival digno de su talento.
"Twin Star Exorcists" narra la historia de estos dos talentosos exorcistas, destinados a convertirse en las "Estrellas Gemelas Exorcistas". Se profetiza que serán los padres del Miko, la reencarnación de Abe no Seimei, quien finalmente liberará al mundo de toda impureza.
